ABSTRACT:Theaimof this study was toreveal the molecular mechanism of lncRNAPCATlcontributing tocell proliferationand migration inpancreaticcancercels.Thereverse transcription-quantitativereal-timepolymerasechainreaction (RT-qPCR)and Western bloting analyses were performed to measure the expression of the indicated genes.The RNA pul-down assaycombined with Western bloting validation,RNA immunoprecipitation(RIP)-qPCR assay,protein stability assy,glucose uptake asy,and lactate production assay were conducted to investigate the molecular mechanism by which PCAT1contributed tothe proliferation and migration in pancreaticcancercels.The results demonstrated that PCATl upregulated the expression of LDHA (lactate dehydrogenase A) in pancreatic cancer cels.PCATl may interact with LDHA protein,but had no efect on its stabilityand degradation in pancreatic cancer cels.Furthermore,PCAT1 promoted glucose uptakeand lactate secretion by up-regulating the expressionof LDHA,therebypromoted cellproliferation and migration. In summary,our results suggested that PCAT1 promoted aerobic glycolysis by up-regulating the expressio of LDHAand interacting with LDHA protein,subsequently promoting the proliferation and migrationof pancreatic cancer cells.
