ABSTRACT:Toanalyzetheexpresion and prognostic significanceof E3ubiquitin ligasering finger proteinl25(RNF125)in livercancerand explore the efectand mechanismof RNF125on invasion and metastasis of livercancer.ENCORIdatabase andclinical tissue samples wereused toanalyze the expression levelsof RNF125inlivercancer andadjacenttissues,and the relationship betweenRNF125and prognosis oflivercancer was analyzed using ENCORIdatabase.Hepatocelularcarcinoma (HCC) cellines with knockdown and overexpression of RNFl25 were constructed,and the effect of RNF125ex- presionlevelon the migration and invasion abilityof HCCcels was examined by Transwell assay and wound healing assay.The efects of regulating RNF125on tumor growth and tumor immune microenvironment were verified by animal experiments.The target proteinof RNF125 was found by mass spectrometry.Therelationship between RNF125and MYH9 was verified byqPCR,Western blot and ubiquitination.The efects of regulationof RNF125and MYH9 on EMT marker proteins were determined by Western blot and celular immunofluorescence techniques.The results showed that the expressionof RNF125 was low in liver cancer tissues,and the high expressionof RNF125 suggested a god prognosis.TheexpressionlevelofRNF125 was negativelycorrelated withthe migrationand invasionabilityof HCCcells.Over expression of RNF125delayed tumor growth and increased the levelofimmunecels invasion.RNF125 specificall bound to MYH9 and promoted its ubiquitination degradation.RNF125 was positivelycorelated with theexpresionlevelof epithelial cell markerand negativelycorelated withmesenchymalcellmarkers.OverexpressionofMYH9 reversedthefect of RNF125 on EMT marker proteins.Inconclusion,the expresion of RNF125 was down-regulated in liver cancer,and the high expresion of RNF125 was related to the good prognosis of liver cancer patients.RNF125 inhibits the EMT process of liver cancer by ubiquitinating MYH9,thus inhibiting the migration and invasion of liver cancer.
