【Abstract】 Objective To investigate the expression characteristics of NLRX1 in lung invasive adenocarcinomaand its correlations with clinicopathological parametersand tumor proliferativeactivity.MethodsA total of 128 surgically resected lung tumor specimens were included.NLRX1 protein expression was detected by immunohistochemistry (IHC),and mRNA levels were quantified by qRT-PCR.The relationships between NLRX1 expression and tumor differentiation degree,Ki-67 proliferation index,lymph node metastasis,andother clinico-pathological factors were analyzed.Results ① NLRX1 protein expression exhibited significant tissue-type speci-ficity: It was negative in benign tumors (bronchiolar adenoma),precancerous lesions,and non-adenocarcinoma subtypes, while showing positive expression in lung invasive adenocarcinoma (29/65,44.6 % ) ② Both NLRX1 pro-tein and mRNA expression demonstrated tumor differentiation-dependent patterns.Protein positivityrates:well-differentiated (80.0%)> moderately-differentiated (47.4%) poorly-differentiated groups (25.8 % ) χ2 =12.097, P =0.002). mRNA levels in lung invasiveadenocarcinoma (1.54±O.75)were significantlyhigher than inadjacent tis-sues(1.00 ± (20 0.26) (t=-4.900, P <0.001),gradually decreasing with reduced differentiation ( F =7.735, P=0.001). ③ Negative NLRX1 expression strongly correlated with: High Ki-67 proliferation index (10o.0% vs. 46.3%, χ2 =8.604, P =0.003). Lymph node metastasis( 83.3% vs. 49.1 % , =4.652, P =0.031).ConclusionNLRX1 may function as a tumor suppressor in lung invasive adenocarcinoma byinhibiting proliferation and metastasis.Its expression level could serve as a potential biomarker for evaluating tumor differentiation status and malignant phenotypes.
