【Abstract】 ObjectiveTo investigate the effects of different baking time and temperature on immunohistochemical staining of electroresectionof prostate.MethodsAtotalof 4O patients with prostate cancer whoseclinicopathological data wererelativelycompleteandarchived intheFirstPeople'sHospitalof Jintan from January 2O21 to January 2O25 were collected.allpatients were diagnosed as malignant and diagnosed as prostate cancer.The samples were placed under different baking time and different baking temperature for immunohistochemical staining of 34βE12，P504sand P63 proteins.ResultsWhen the tissue slices were placedinthe electric thermostatic drying oven at 55～6O℃，the baking time was set as1h， 4h ，16hand24hrespectively，indicating that theantibody immunostaining intensitydid not change significantlyand wasnot affected significantly.However，it wasfound thattheimmunostaining intensityofbaking slicesdecreased gradualywiththechangeofbaking timeafter 6hin the electric thermostaticdrying ovenat75～8OC.Inthis study，itwas confirmedthat the prostate tissue slices werebakedat55～6O℃forlessthan16hwithoutafecting the preservationof mostantigenactivity.Using 34βE12， P504sandP63 proteins as target antigens toanalyze the immunohistochemical staining of electroresectionof prostate has positive clinical application value.ConclusionImmunohistochemical staining is a relatively complex process， and the increase ordecrease of antigens may occur in the whole processfrom the preparation of tissue materials for immunohistochemical examination to the final immunostaining.
