【Abstract】 Objective To investigate the expressionof Toll-like receptor 4（TLR-4），interleukin-12（IL-12） andcyclooxygenase-2（COX-2）incervical cancer tissues andanalyze their relationshipwithhigh-risk human papillma virus （HPV） infection.MethodsFrom March 2021 to November 2024，We collcted Zhengzhou Central HospitalAfiliated to Zhengzhou University118 patients withcervicalcancer，112patients with cervical intraepithelial neoplasia（CIN）and1O6 patients with chronic cervicitis（CCS）were divided into cervical cancer group，CIN groupand CCS group.The expressonof TLR-4，IL-12，COX-2 and high-risk HPV infection incervical tissues of three groups and CIN group were compared.To compare the expression of TLR-4，IL-12 and COX-2 in cervical cancer tissues with diferent HPV viral loads，and to analyze the correlation between high-risk HPV infectionand the expresion of TLR-4，IL-12 and COX-2 incervical cancer patientsbySpearman correlation analysis.ResultsTherelative expressonof TLR-4，COX-2and thepositive rateof high-risk HPVincervical cancer group were higher than those in CIN group and CCS group，and CIN group was higher than CCS group（ P< 0.05）.The relative expresson of IL-12 in cervical cancer group was lower than that in CIN and CCS groups，and CIN group was lower than CCS group（ P <0.05）.The relative expression of TLR-4 and COX-2 and the positive rate of highrisk HPV in different grades of CIN patients were as follows：CIN III > CINII > CINI（ P <0.05）. The relative expression of IL-12 in different grades of CIN group was as follows： CIN II HPVload grade 2>HPVload grade 1（ P <0.05）. Spearman analysis showed that high- risk HPV infection was positively correlated with the expression of TLR-4（ r= -0.547，P<0.001） and COX- 2（r=0.649，P<0.001）， and negatively correlated with the expression of IL-12 in cervical cancer tissues （ r=0.745 ，P<0.0o1）.Conclusion The expressions of TLR-4，IL-12 and COX-2are closelyrelated to the progre-sionofcervi-cal lesions and cervical cancer，and areclosely relatedtohigh-riskHPVinfection.
