[Abstract]Periodontitis isanimmune-inflammatoryresponse triggeredbyperiodontalpathogens.Inthiscondition,disruptedimmune responses and chronic inflammation play crucial roles in the degradationof periodontal issues.Cuproptosis,arecentlyidentifiedforofprogammedcelldath,istriggeredbyexcessiveintracellularcoperionsandspecificallytargets thetricarboxylicacidcycle.Itischaracterized bytheabnormaloligomerizationof mitochondrial lipoacylated proteins and the loss ofiron-sulfur cluster proteins.Cuproptosis has garnered significantatention for its nonnegligible role inthedevelopmentand progression of variousdiseases,including cardiovascular diseases,neurodegenerativedisorders,and cancers.Cuproptosis-related genes participate inthepathogenesis of periodontitis bymodulating immune cells and cytokines.The abnormal expresionofthese genes reflects the disease status andaids in clinicaldiagnosis,providing noveltherapeutic targets for periodontitis treatment.Considering these insights,this work presents a comprehensivereviewof the molecular mechanisms underlying cuproptosis and itsrole in the initiationand progression of periodontitis.
