[Abstract]ObjectiveThis study aimed to assessthe causal relationship between programmed celldeath ligand 1 (PDL1)and the riskof head and neckcancer.MethodsMendelianrandomization (MR)was performedusing large-scale geneticdatatoevaluatethecausalrelationshipbetweenPD-L1andtheriskofdiferent typesofheadandneckcancers via inverse variance weighted (IVW),Mendelian randomization-Egger (MR-Egger),simple mode,and weighted median. Additionaly, MR-Egger and Mendelian randomization pleiotropy resiual sum andoutlier (MR-PRESSO) were used to analyze horizontal pleiotropy,Cochran’sQtest was performed toexamine heterogeneityand leave-one-out experimentwas conducted to measure sensitivity.ResultsThe genetic prediction results revealeda causal relationship between PD-L1 and head and neck squamous cell carcinoma (HNSCC) (OH R=1.000 3 , 95%Cl :1.000 0-1.000 6, P=0.049 )andbetween PD-L1 and oral cavity and oropharyngeal cancer (OC&PC) (OF :=1.000 3 95% CI: 1.000 1-1.000 6, P=0.018 ). Conclusion PD-L1 may be a risk factor for the development of HNSCC and OC&PC.
