Comparative Study of Different Cryopreservation Systems for Chicken Primordial Germ Cells
SUNShengrui ,ZHUYing,ZHAOHaoyi,HEYanhua,ZHAO Zhifeng, LUO Chenglong, SHU Dingming, ZOU Xian (1.IInstituteofAnimalScience,Guangdong AcademyofAgricultural Sciences & StateKeyLaboratoryofSuineand Poultry Breeding Industry& Key Laboratoryof Animal Nutrition andFeed Sciencein South China,Ministryof Agriculture and Rural Affairs,Guangzhou Guangdong 510640; (2.Huazhong Agricultural University,Wuhan Hubei 430000) Abstract:To identify the optimal cryopreservation protocol for chicken primordial germ cels (PGCs),eight cryopreservation solutions-including both literature-reportedand commerciallyavailable formulations-were evaluated.Two differentcellconcentrations werecryopreserved under identical culture conditionsand processing method.Post-thawanalyses,including trypan blue exclusion assay,CCK-8cell proliferation assay,andapoptosis detectio were used toperformed toassess cellviabilityand status in each group,thereby evaluating theperformance of the eight cryoprotectantsateach cellconcentration.The results showed thatt Cryoprotectant7yielded the highest number of viable cells at a concentration of 1.37 × 1 06 cells/mL,reaching 1.14×106 ,with 74.8% of cell being negative forbothfluorescein isothiocyanate(FITC)and propidium iodide(PI).Incontrast,Cryoprotectant1yielded the highest number of viablecellsat 2.75×106cells/mL,reaching2.60×106,with 75.2% ouble-negative cells.However, Cryoprotectants II and M showed the least favorable outcomes.These findings indicate that the effectiveness of cryoprotectants varies with cell concentration.The most suitable solution for cryopreserving PGCs at 1.37 × 1 06 cells/mL in 250μL was Solution M (Saichang Biological Solution).For cryopreservation solution PGCs at 2 .75×106 cells/mL, the optimal formulation was SolutionI (basal medium:chicken serum:DMSO CaCl=81.25%:10%:8%:0.75%).
