Abstract:An effective method was developed for the determination of acrylamide in tea by liquid chromatography-tandem mass spectrometry with dispersive solid-phase extraction combined. The sample was soaked in water and extracted with pure acetonitrile, and purified by 900mgMgSO4 300mg PSA, 300mg (204号 C18, 90mg GCB, 300mg SiLica. After separation on a XSELECT HSS T3 (2.5μm, 3.0mm×100 (20号 mm)column, the analytes were detected by LC-MS/MS in the multiple reaction monitoring(MRM) mode and quantified by internal standard method of solvent standard curve. Good linearity was obtained for acrylamide in the linear range of 0.5-250.0μg/L ,with correlation coefficient( r2 )larger than O.999,The limit of quantitation(LOQ) was 1.0μg/kg .The average recoveries at three levels of 10,20, 50μg/kg ranged from 90.4% to 101.3% ,respectively. The relative standard deviations(RSDs, n=6 )ranged from 0.9% to 3.3% .280 samples were detected and 244 samples were found to contain acrylamide with a concentration ranging from ND to 305.7μg/kg .However, this method have certain advantageous such as simple operation, high sensitivity, and good accuracy.In conclusion,this method is suitable for the determination of acrylamide in tea.The assessment of the exposure risk of acrylamide in tea using the exposure limit method (MOE) indicates that there may be a high exposure risk under extreme ingestion conditions, which should be of concern to relevant departments.
