Abstract: The SH-related sequence(SS)gene family encodes aclass of plant-specific zincfinger protein transcriptionfactors hat playcrucial roles inregulating plant growth,developmentandstressresponses.Toexplorethefunctionofte SRSgenefamily n tobacco,18NSRSgeneswereidentifiedfromthetobacogenomedatabaseanddesignatedasNSRS1toNtSRS18.Bioinformatics analyseswereconductedtoinvestigatetheirproteincharacteristics,subcelularlocalization,phylogeneticrelationships,gene structures,andooterentditoallyprioaesoilysdeenttisssdsel theirresponses todrought,salt,andlowtemperaturestresswereanalyzedusingqRT-CR.Resultsrevealedthat NtSRSproteins containedtheconservedDUF702domain,withmostlocalizedin the nucleus.Phylogeneticanalysis clusteredtheNtSRSfamilyinto 5 subfamilies,with membersofthesamesubfamilyexhibitinghighlyconservedgeneand protein structures.Promoteranalysis identifiednumeroushormone-responsiveelementsand stres-responsive elementswithintheregulatoryregionsof NtSRSgenes. Expresion proilingshowed thatNtSRSgnes exhbitedsgnificant tssuespeciic expresson,withosthighlyexpressd inflower budsand stems.NotablyNSRS3and NSRS16 were stronglyinducedunderdrought,salt,andlowtemperature strescoditios. OverallthisstudyprovidesacomprehensivecharacterzationofthetobaccoSRSgenefamilyofferingatheoreticalbasisforthe genetic improvement of tobacco germplasm resources.
