Comparative Analysis of Rhizosphere Microecology Between Healthy and Root-Knot Nematode-Infected Tobacco Plants

Abstract: Inorder toanalyzetherelationshipbetwenrhzosphere soilmicroecologicalfactorsandtheoccurrenceoftobaccorootknotnematodedisease,atypicaltobaccofieldwithtobaccoroot-knotnematodediseasewasselectedinAziingTownship,Songing County,KunmingCity,Yunnan Province.Rizospheresoilsamples werecolected fromboth diseasedandhealthytobaccoplants. The numberofsecond-istarlarvaeofot-knotnematodes,soilchemicalproperties,enzymeactivities,andmicrobalcommunity structure were analyzed andcompared.Results showed thattherot-knot nematode pathogen was identified as Meloidogyne incognita.Comparedwiththerhzospheresoilofhealthytobaccoplants,thenumberofroot-knotnematodesintherhizospheresoilof diseased tobacco plants increasedby4.29times ( p<0 . 0 1 ) .Soil acid phosphatase activity,pH,and available phosphoruscontent were reduced by 8 6 . 9 8 % ( p<0 . 0 5 ) , 0.33 units ( p<0 . 0 1 ) 4 5 . 5 3 % ( p<0 . 0 5 ) ,respectively. In contrast, sucrase activity increased by 5 7 . 3 6 % (204 ( p<0 . 0 5 ) .The microbialbalance,bacterialdiversityandrichness decreased,andthefungaldiversityincreasedbuttherichess decreased.ThebacterialgenusSphingomonaswereenrichedintherizospheresoilofthediseasedtobaccoplants,andthefungal phylumAscomycota wereenriched intherhizosphereofthehealthytobacoplants.Thenumberof2ndinstarlarvaeofroot-knot nematodewasignicantlypositivelyoeatedwithucraseactivityndSpngomonasndigncantlyegatielyeadih pH,acid phosphatase,availablephosphorusandAscomycotaLowsoilpHacid phosphataseactivityandavailablephosphorus content,and high sucrase activity may be important factors in inducing tobacco root-knot nematode disease.
