Abstract:Tofurtheroptimizethemicroenvironmentparametersofsugar-free tisseculture,this studyused Qingshu No.9 as the tst material and designed different gradient experiments with diferentratiosof vermiculite to nutrient solution ( Vvermiculite : 1 Vnutrientsolution=1.0:0.7,Vvemiculie:Vnutrientsolution=1.0:0.8 mtriensoution =1.0:0.9),COconcentrtions(500 μmol/mol, 800μmol/mol , 1100μmol/mol , 1 400μmol/mol ),and light intensities to investigate the effectsof each treatment

onthe growth and development of potato tissue-cultured seedlings.Thelight intensitywasset at 50μmol/(m2⋅s) 75μmol/(m2⋅s) , 100μmol/(μ2⋅s). The results showed that the optimal conditions for sugar-free tissue culture of potatoes were as follows : , CO2 concentration of 800μmol/mol ,and light intensityof75 μmol/(m2⋅s). Compared with traditional tissue culture,the yield,quantity,and survival rate of potato pre-basic seeds obtained through sugar-free tissue culture were significantly increased by 16.09% , 22.37% ,and 3.27% ,respectively( P<

0.O5).Inconclusion,thisstudynotonlyclarifidthekeytechnicalparametersofsugar-freetissueculturebutalsoonfimedthe advantagesoftistechologyinimprovingteproductionefiencyofpotatoseedtubers,providingteoreticalbasisadtecal support for the standardized production and industrial propagationof potato tissue-cultured seedlings.
