Establishment of Tissue Culture System for Populus simonii ×P . nigra BasedonStemBurial Rejuvenation of Adult Elite Trees

Abstract To establish a tissue culture system for elite trees of Populus simonii ×P . nigra via stem burial rejuvenation,semi-lignified branches from cuttings of rejuvenated adult elite trees of Populus simoni ×P . nigra after stem burial treatment were used as explants.The optimalexplant disinfection method was screened,and conditions suitable for callus induction from Populus simonii ×P . nigra leaves were explored,aiming to establish an efficient and stable tissue culture system for Populus simonii ×P . nigra .The results showed as the following four aspects. (1)The combination of 75% alcohol disinfection for l2O seconds and 0.1% (20 HgCl2 disinfection for7 minutes had the best effect,with a contamination rate of 17.33% .(2)The optimal medium formula for leaf callus induction of Poϕ 1 ulus simonii ×P . nigra was M 1S+0.05mg ⋅L-1NAA+0.5mg L-16-BA+30g ⋅L-1 sucrose,under which the induction rate was 70% .(3) In the proliferation stage,the optimal medium formula was MS+O.O5 mg L-1NAA+30 (204号 g L-1 sucrose,with a proliferation coefficient of 8.32.(4) The optimal seedling strengthening medium for tissue culture seedlings of Populus simonii ×P . nigra was 1/2MS+0.01mg L-1NAA+0.05mg L-1IBA+20g L-1 sucrose,and the growth amount under this culture condition reached 9.37cm
