Cloning and Expression Analysis of PmSYP8/ Gene from Picea mongolica in Sandy Land

Abstract To explore the molecular structural characteristics of the PmSYP8l gene from Picea mongolica and its relative expresion levels in diffrent tissues,Picea mongolica was used as the material.Total RNA was extracted, and the cDNA sequence of the PmSYP8l gene was amplified by RT-PCR.This sequence was cloned into the pMD19-Tvector,and after sequencing verification,bioinformaticsanalysis were performed,including amino acid sequence deduction,analysis of physical and chemical properties of the protein,prediction of secondaryand tertiary structures,and homology alignment. The results showed that the full-length of the PmSYP81 gene was 942 bp,encoding 3l4 amino acids.This protein has one N-terminal domain and one SNARE superfamily domain,and shares high homology with seven plant species including Arabidopsis thaliana,Helianthus annuus,and Platanthera zijinensis.PmSYP81 protein has a transmembrane structure and is a hydrophilic protein.The proportion of serine and threonine sites is high.The PmSYP81 gene was sucessully cloned and analyzed in many aspects,which laid a foundation for in-depth study of the biological function of this gene in Picea mongolica.
