Determination of Cembratriene-diols in Tobacco by Using Heart-cutting Two-dimensional Liquid Chromatography Method

Abstract:To solve theproblemsof lowcontent,numerous interferences,and dificulty inaccurate determinationof cembratriene-diols(CBD),the importantflavorprecursors intobaccoleaves,a heart-cutting two-dimensional liquid chromatography method was established for the efficient determination of α -CBD and β -CBD contents in tobacco leaves. A heart-cuting strategy was used to remove most interfering components,and the sample pretreatment was simple.The proposed method only extracted the target CBD from tobacco leaf powder with ethylacetate,followed bycentrifugal separationand filtration before being testedonthe instrument.Theresultsshowthat thismethod exhibits high sensitivityand good reproducibility,with the square of linear correlation coefficient R2 greater than 0. 999,recovery rates exceeding (204号 93% ,and intra-day and inter-day repeatability both below 5% . The mass concentration limits of quantification for α- (204号 CBD and β -CBD are 1.10 mg/L and 0.80mg/L ,respectively. The CBD content in tobacco leaves is significantly influencedby growing regionandvarietybutlessaffectedbyleaf positionandnitrogenaplication.Forfreshtobaccoleaves of the sameoriginandvariety,theCBDcontentissignificantlyhigherthanthatincuredleaves,highlightingtheneedtocontrol CBD degradation during the curing process.
