Abstract:Objective To establish an ultra-performance liquid chromatography-tandem massspectrometry(UPLCMS/MS)method for simultaneous quantificationof 12 active components in Shenling Baizhu Granules.Methods Chromatographic separation was performed on a Waters Atlantis T3 column ( 4.6mm×75mm , 3μm )at 30∘C , with a mobile phase consisting of 0.1% formic acid in water and methanol under gradient elution.The flow rate was (202 0.4mL⋅min-1 ,and an electrospray ionization (ESI) source was employed for detection in both positive and negative ion multiplereaction monitoring(MRM) modes.Results The 12 active components [platycodin D,chikusetsusaponin Na , pseudoginsenoside F11 ,allantoin,liquiritin,atractylenolideI,atractylenolide II ,atractylenolide Il ,ginsenoside Rf,ginsenoside Rb1 ,ginsenoside Re,and ginsenoside Rgil exhibited good linearity ( r>0.9977 )within their respective concentration ranges.The average recoveries ranged from 96.16% to 102.13% with relative standard deviations(RSD) <4% .Significant variations in the content of these 12 components were observed across eight batches of Shenling Baizhu Granules.Conclusion This method is simple,accurate,and reproducible,providing areliable approach for quality control and research of Shenling Baizhu Granules.
