Abstract:Objective To investigate the corelation between the content of six conjugated anthraquinonesand the purgative ffcts of Rheum palmatum from diferent origins.Methods The contents of aloe-emodin-8- o -glucoside (A8G),rhein-8-O-glucoside(R8G),chrysophanol-8-O-glucoside(C8G),emodin-8-O-glucoside(E8G), physcion-8- υ -glucoside(P8G),and sennoside A in Rhei Radix et Rhizoma from 1O geographical origins were determined using high-performance liquidchromatography(HPLC).The purgative efect of Rhei Radix et Rhizoma decoction onnormal mice were evaluated based on purgative index and charcoalpropulsion rate.Grey relational analysis and corelation analysis wereemployed to explore therelationship between thecontent of six conjugatedanthraquinones andpurgative fects.Results Statisticallysignificant differences wereobserved inthecontent of sixconjugated anthraquinones among the 1O origins ( P<0.05 ).Rhei Radix et Rhizoma from Enshi,Hubei Province,exhibited the highest levels of A8G,R8G,C8G,P8G,and sennoside A,while samples from Haiyuan,Ningxia,showed the highestE8Gcontent.Significant variations in purgative efects(purgative index and charcoal propulsion rate)were observed across origins,with Rhei Radix et Rhizoma from Enshiand Jianshui demonstrating the strongest effects ( P< 0.05).Greyrelationaland correlationanalyses revealed apositive corelationbetweenconjugatedanthraquinones content andpurgative effects.A8G,C8G,P8G,and sennoside A contributed significantly to both purgative index and charcoal propulsionrate.Conclusion The purgativeeffects ofRhei Radix et Rhizoma arises from thesynergistic effects of multiplecomponents,with A8G,C8G,P8G,and sennoside A likelyserving as key active constituents.These findings provide a foundation for further exploration of the purgative mechanism of Rhei Radix et Rhizoma.
