Abstract:【Objective】TheaimofthisstudywastocloneLdCSP1andLdCSP2genesofchemicalreceptorproteinsofLymantria dispar,andtostudythechaacteristicsgeexpressonandesstaetodugstressexpressonofdCSPsgnes.histudylaid foundationforrevealingthecharacteristicsofchemicalreceptorproteinsLdCSP1andLdCSP2genesandtheirspecificfunctionsin drugmetabolism,andalsoprovidedanewideaforfiding potentialdrugtarget genesand preventinggypsymoth.【Method】Based onthetranscriptomesequencingresultsofLdispar,twochemicalreceptorproteingenesofLdisparwereconedbyRT-CRORF Finder,ExPASyignalP4.1NCBICDandtheronlinetoolswereusedforoiformaticsanalysis.qR-PCRwasusedtoaalye theexpressionofCSPgenesindiferentspeciesofL.disparanduderrotenonestress.【Result】ThetotallengthofLdCSPgeneORF is360bp,encoding119aminoacids.hetotallengthofLdCSPgneORFis375bpencodng124aminoacids.Sequencealsis showedthattheaminoacidsequencesof thetwoCSPofLdisparhad4conservedcysteineloci,nd wereconsistent withtheCSPs structurepatern“C1-X6-C2-X18-C3-2-C4.PhylogenetictreeanalysisshowedthattwoLdCSPswereclusteredintotwobraches ofotherepidopteraCSPs,andeachLdCSPsequencewasclusteredintoasmallbranchwiththesequencewithhigherhomologyThe expressionofLdCSPgenesatevelopmentstageshowedthatLdCSPgeneswereexpressdinallstagesofL.disparandterelative expressionlevelsofLdCSPandLdCSP2genes werethe highestathethirdstage (I3D1),andthelowestattheeggstage.Tissue expression showed that LdCSPs wereexpressed in different tissues of L disparatthe thirdinstarlarvaestage. LdCSPs genes had the highest expression in the thorax,and the tissue expression of the two LdCSPs was different.QRT-PCR results showed that the expression ofLdCSPsgeneinththidinstarlarvaeofL.disparwasfstdecreasedandthenup-regulatedunderrotenoestres.【Concusion】Two Chemosensory proteingenes,LdCSP1andLdCPS2,wereclonedfromL.dispar.QRT-PCR showed thatLdCSPs gene expressionhad timeand dose effects on rotenone stress,indicatingthat LdCSPs playedan important role in themechanism of detoxification resistance ofL.dispar,whichprovidedareferenceforfurtherstudyofthefunctionofthesedrugtargetgenes,ndprovidedanewideaforfuture preventionand control ofL.dispar.
