Abstract:【Objective】This studyclonedthePkSERK genes from Korean pine(Pinus koraiensis)andanalyzedtheir expresion characteristics,layingafoundationforfunctionalresearchonPkSERKsinsomaticembryogenesisofKoreanpine.【Method】Tefullengthcodingsquence(CD)ofERK-3eneinKoreanpineassuessfullloed,subsequentbifomaticsalysere conducted toexaminetheproteinstructure,pyicochemicalpropetis,andevolutionaryelationships,subcellarlocalationwas observedithotetalldps koreanpinecalliwereanalyedy-CRtechnology【Result】SERK1,SERK2,andSERK3weresuccessullyod,i cDNAlengthsof1824,1887and1437bp,encoding 607,628and448amnoacids,respectively.PkSERK1isanacidic,hdohlic andstableprotein,hileERK2isanacidic,hydrophilic,andustableprotein,ndPERK3isbasic,hydrophilicndstable protein;bothPkSERKlandPkSERK2containasignalpeptide,whereasPSERK3doesnothaveasignalpeptide;tesetrproteins arealltransmembraneproteins.ThePkERKlproteinhastheclosestgeneticrelationshipwithCsERK1-3ofCuninghamialanceolata,

PkSERK2hastheclosestgeneticrelatioshipwithCsSERKlofCameliasinensisandPkSERK3hastheclosestgeneticrelatiship with CjSERK3ofCryptomeriajaponica.PkSERK1andPkSERK2werelocatedinthecellmembrane,whilePkSERK3waslocatedin thecellmembraneandnucleus.TePkERK1geneashighlyexpresedinbothcalluswithighembryogenicpotentialandcaluswith globularembryosofkoreanpie;PkSERK2washighlyexpressedinbrowncalls;PkSERK3wasonlyhighlyexpressdincallus with highembryogenicpotential.【Conclusion】TegenesofPkSERK1,PkSERK2andPkSERK3wereclonedfromkoreanpine,PkERK1 mayregulatethemaintenanceofembryogenicpotentialincallusandearlyglobularembryodiferentiationinKoreanpine,high expresionofPkSERK2maybeassciatedwithbrowningembryogeniccalus(EC)andearlyglobularembryodiferentiationinKorean pine,while PkSERK3 may onlyplaya role in the maintenance of embryogenic potential in Korean pine callus.
