Abstract:To establishan eficientvirus-induced gene silencing(VIGS)system in important Brasicaceae vegetables,Brassca rapa subsp.chinensisand Brassica juncea were chosenas the material and theendogenous phytoene desaturase ( PDS )gene was selected as the marker gene.The pTRV2-BrPDS,pTRV2-BjuPDS- and pTRV2-BjuPDS- c vectors were constructed,and the infection efciency of Agrobacterium was optimized to construst VIGS system with high efficient.The results showed that the pTRV2-BrPDS,pTRV2-BjuPDS- g and pTRV2-BjuPDS(204号 c vectorswere successfully constructed.The albinism phenotypeof B.rapa subsp.chinensis plants was the most obviouswith OD 600 =1.0 of Agrobacterium,while the silencing efficiency ofB.juncea wasoptimal with OD 600 =0.8of Agrobacterium.TheqRT-PCRanalysisconfirmedthattheobserved albinismphenotype wasresulted fromthesilencing of theendogenousPDSgene inplant infectedwith the TRV-basedrecombinant virus.Aboveresultsprovided theoretical basis for studing the gene functions of B.rapa subsp.chinensis and B. juncea using VIGS technology.
