Screeningand Identificationofa Strain of Bacillusvelezensis AgainstBotrytiscinerea

Abstract:InordertoisolateabiocontrolbacteriawithantagonsticfetsonBotrtiscinerea,theantagonisticbacteriaere screned from graperhizosphere soilsbyplateconfrontation methodandliter paper method.Astrain with strong antagonistic efectwasselectedandidentifiedbymorpholoigical,physiologicalandbiochemicalcharacteristics,16SrDNAsequences analysis.Thesafety,growth-promotingand thecontrol efectof thestrain were testedbytheblood platemethod,seed germination experimentsanddetached fruitmethod.Theresultsshowedthat5antagonisticbacteriawerescreened from142 bacterialstrainsisolatedfromgraperhizospheresoils.These5antagonisticbacteriahaddiferentinhibitoryefectongrape gray mold disease with the inhibition rates from 81.74% to 89.13% ,and the inhibition zone were from 2.42 to 11.08 mm, amongwhichtheinhibitoryeffectof strainB1-4wasthehighest.Bymorph-oloigical,physiologicalandbiochemical characteristics,16SrDNA sequences,strain B1-4 was identifiedasBacilusvelezensis,which hadno hemolysis,could producecelulase,proteaseand siderophores,and hadnitrogen fixation efect.Thegerminationrateandradiclelengthof cucumberandrapeseed seedscould besignificantly improved by10O times dilutionofB1-4fermentationsolution.The protective effect of B1-4 fermentation liquid reached 70.31% and the rapeutic effect was 69.53% on postharvest grape fruit gray mold,which was no significant diference with 5 OoO times dilution of 50% fludioxonil ( 71.88% ).Above results showed thatB. velezensis B1-4 was a protential biocontrol strain for control B. cinerea.
