延迟激活及不同处理方式对猪体细胞克隆胚胎体外、体内发育效率的影响

Abstract:The literature shows that electrofusion parameters,fusion methods and treatment methods of cloned embryosmayafecttheinvitrodevelopmentefficiencyofporcinecloned embryos.Theeffctsofsynchronousactivation anddelayedactivationoninvitroandinvivodevelopment eficiencyof porcinecell cloned embryos werecompared. All embryos were placed in PZM-3 culture medium containing 5μg/mL of cytochalasin B(CB) for 4h after activation. There was no significant difference in the fusion rate between the two groups( P>0.05 ),but the cleavage rate and blastocyst development rate of delayed activation were higher than those of synchronous activation group.CB and histonedeacetylase inhibitor arealsocommonlyused inthe treatment ofactivated cloned embryos.This study also comparedthe treatmentwithcytorelaxinBafterdelayedactivationandthetreatmentwith histonedeacetylaseinhibitor scriptaid,andfoundthatthelatter was moreconduciveto improving theefficiencyofinvitro embryodevelopment.The cloned embryosobtainedbytheabovedifferent methods were transferred,andthe statisticalresultsshowedthat the delayed activation method was moreconducive to embryodevelopment.After delayedactivation,CBor scriptaid was usedtotreat theembryos,andthepregnancyrateand cloning eficiencyaftertransplantation were significantlyhigher than those in the synchronous activation group.
