2,3,7,8-四氯二苯并二嘌英通过miR-214-3p抑制间充质细胞的增殖和迁移导致聘裂的形成

【Abstract】Objective:To investigate the role of miRNA(miR)-214-3p in a cleft palate induced by 2,3,7,8 -tetrachlorodibenzo-pdioxin(TCDD).Methods: We establisheda cleft palate model by intragastrically administering TCDD ( 64|μg/kg )to pregnant C57 mice.The expressionof miR-214-3p in mouse embryonic palatal mesenchymal(MEPM)celswas measured by miRNA sequencing andquantitativereal-timepolymerasechaireaction(qRT-PCR).WeexamiedtheefectsofTCDDviamiR-214-3pontheprolferation and migration of MEPM cells by using Cell Counting Kit- 8,5- ethynyl- 2 '-deoxyuridine assay,transwell assay,and wound healing assay.Results:Exposing C57 mice at 10.5 days of gestation to TCDD(64 μg/kg),32 of 32 fetuses( 100% ) in the experimental group developedacleftpalate.Nocleftpalateoccurredinthecontrolgroup(O/28).TheTCDDgroupshowedsignificantlyup-regulatedexpressionofmiR-214-3p,miR-296-5p,andmiR-3-5pandsignificantlydown-regulatedexpresionofmiR-92a-3p,miR-126a-3p, and miR-411-5p.TheqRT-PCRresultsverifedmiR-214-3passignificantlydiferentiallyexpressed.ThemiR-214-3pihibitor partiallyrestoredteproliferationandmigrationabilitiesofEPMcellexpoedtoTCD.Conclusions:TCDDcansuppresthepro liferationand migrationof MEPMcelsthrough miR-214-3p,indicating thatmiR-214-3pmayplayacrucialrole in TCDD-induced cleft palate in mice.
