【Abstract]Objective:To investigate theexpressionofFERMT1inlungadenocarcinomacellsandtheeffectand mechanismof FERMT1ontheproliferatio,vasionigration,ndapoptosisoflungadenocarioacelsbygulatingtheWntsigalingpathay. Methods:ThebioinformaticsanalysiswasusedtoinvestigatetheexpressonofFERM1inlungadenocarcinomaanditsassociation withclinicalfeaturesndprogosis;initroexperimentsreuedtoivestigateteifuenceofFERontiologicalavior of lung adenocarcioma;qRT-PCRandWesteblotwereusedtoobserve theefectofthechangeiFERMT1expresiononthe Wnt/β -cateninpathwayanditsdownstreamtargetgenes.Results:ThebioinformaticsanalysisshowedthehighexpresionofFERinlng adenocarcinomatissue,theexpressonlevelofFERMTinTNMstagingoflungadenocarcinomawashigherthanthatinnomalung tisue,andmeanhile,theighexpresioofFERledtotheoorprognosisoflungadenocarciomapatients,ainlythoughthe Wntsignalingpathway.InvitroxperimentsshowedthatibiionofFERMexpresioneducedtheproliferaton,migrationd invasionabilitiesoflungadenocarcinomacellsandincreasedtheapoptosisoflungadenocarcinomacells.TheresultsofqRTPCRand Western blot showed that the change in FERMT1 expression affected the expression of β- catenin,epithelial-mesenchymal transition-related markers,and their downstream target genes. Conclusion:FERMT1iscloselyassociated with the development andprogressionoflungadenocarcinoma through theWnt/β-catenin signaling pathway.
