Thymol improves the core symptoms of autism through regulatingscinderin expression

MethodsTheASD-likebehaviorsofratswereoservedusingthreeboxsocialexperiment,penfieldexperimentandyouthsoialexperiment.RNA-seqwasusedtodetecttheexpresionofRNAintheprefrontalcortexofrats.Theproteinlevelofscinderinntheprefrontalcortexofratswas measuredbyWesternblot.Results:Comparedwith therats inthecontrolgroup,theratsinthemodelgroup showedAlisdllidialesdloiilit stereotyped behavior.Compared with the control group,the model group showed reduced standing times( P<0.001 )and increased self combing time( P =0.003).Moreover,theexpresionlevelofscinderinintheprefrontalcortexincreasedinmodelrats.Afterthymolreatment,theASD-likebehaviorsinthe modelgroup weresignificantlyimproved.Comparedwiththe model group,thenumberofstanding times in the intervention group increased( P<0.001 )and the self grooming time decreased( P=0.004 ).The expression of scinderin basically returnedtothenorallevel.Conclusion:TheabnoalexpressonofsinderinintheprefrontalcortexofASmodelatscan lead to ASD-likebehaviors.Thymol interventioncanreducethe expresionofscinderinandimprovethecore symptoms of ASD. 【Key words】autism spectrum disorder;valproic acid;thymol;scinderin
