ABSTRACTOBJECTIVETo explore theimprovement efectand mechanismof salidrosideonradiation-induced parotid gland injury inrats.METHoDsRats wererandomly asigned into normal group,radiation group,salidroside low-dose(salidroside-L, 50mg/kg )group,salidroside high-dose(salidroside-H, 100mg/kg )group,and salidroside-H+inhibitor( 100mg/kg salidroside +0.1 μmol/kgH-θ9 )group,with10 rats in each group.Except forthe normal group,rats in the other groups were subjected to radiation exposure toestablishamodelofradiation-inducedparotidglandijury.Rats ineachgroupwereintraperitoneallinjectedwiththe corresponding drugornormal saline,onceaday,for40consecutivedays.After thelastadministration,thelevelsofeactive oxygen species(ROS),cyclic adenosinemonophosphate(cAMP),superoxide dismutase(SOD),and amylase in serum were detected;thepathological changes of parotidglandtissewereobserved;theapoptosisrateof parotidglandtisuecells,the expressionlevelsofB-cellymphoma-2(Bcl-2)anditsassociatedXprotein(Bax),mRNAexpressionlevelsof interleukin-6(- 6)and tumor necrosis factor ⋅α (TNF- α⋅∝ ),the protein expression levels of type Il collagen((Col II),vasoactive intestinal peptide (VIP),andthephosphorylationlevelof protein kinaseA(PKA)inparotidglandtisse were determined.RESULTSCompared with normal group,thelevelsofROS,amylase,apoptosisrate,Bax expresion level,mRNA expressonlevelsofIL-6andTNFα,and protein expression level of Col II in the radiation groupweresignificantly increased,while thelevelsofcAMP, SOD,Bcl-2 expression level,VIP protein expression level and PKAphosphorylation level were significantly decreased ( P< 0.05).Compared with radiation group,the above indicators in the salidroside-L group and salidroside-H group were significantly improved (P<0.05) ),and the improvement in the salidroside-H group was more significant ( (P<0.05 );inhibitor H-89 significantly reversed the changes in the above indicators of the salidroside-H group( ⋅<0.05 ).CONCLUSIONS Salidrosidecan effectivelyalleviateradiation-induced parotid gland injuryin rats,and its mechanism may be related to the activation of the VIP-cAMP pathway.
