ABSTRACTOBJECTIVETocompare theanti-hepatitismechanismsofAbruspulchellussubsp.cantoniensis(Hance)Verdc (AC)and Abrus pulchelussubsp.molis(Hance)Verdc.(AM).METHODs SDrats were randomly divided into blank group,ACtreated group,andAM-treatedgroup,witheachgroupconsistingof1Orats.Therats’orbitalvenous bloodwascolectedat5,15, 30 minutes,and 1,1.5,2,4,6,8,12 hours after gavage administration of 24g/kg of the corresponding drug (calculated by crudedrug)orwater,respectively.Utra-performanceliquidchromatography-quadrupoletime-of-flightmassspectrometry technologywasutilizedtoidentifytheprototypecomponentspresentintheserum.Thenetworkpharmacologymethodwasadopted topredicttheanti-hepatitisactivecomponents,keytargets,andsignalingpathwaysofACandAM.Aditionally,molecular docking technologywasuilizedtoverifythebindingactivityofthecoreactivecomponentswithkeytargets.RESULTSAtotalof 35prototypecomponents migrating tothe bloodofACandAM wereidentified in theserumofadministeredrats,among which 24 were commoncomponents.The activecomponents inAC,suchasacetylanguidine,physcion,soyasaponinA3andsoyasaponinI,aswell asthose inAM,includingvicenin3,acetylanguidine, soyasaponin I and schaftoside,all acted on key targets such assteroid receptor coactivator, phosphatidylinositol-4,5- bisphosphate 3-kinasecatalyticsubunit alpha, epidermal growth factor receptor(EGFR),and protein kinase B1 (Aktl). These components modulated pathways in cancer, EGFRtyrosinekinaseinhibitorresistance,andthe phosphoinositide3-kinase(PI3K)-Aktpathway,thereby exerting anti-hepatitis effects. Furthermore, thebinding energiesbetween these activecomponents and their key targets were all less than .CONCLUSIONS There are differences in the active components of AC and AM against hepatitis, buttheirmechanismsofactionaresimilar.Bothmayexerttheiranti-hepatiseffectsthroughpathwaysincancer,EGFRtyrosine kinase inhibitor resistance,and the PI3K-Akt pathway.
