Abstract:InordertoimprovethefciencyofproducingliquidspawnofStrophariarugosoaulata,ascreningmethodwasusedto conductatriangularflaskcultureexperimentand9orthogonalexperimentsonthebasicliquidstrainformulasofStrophariarugosoanulatawithdiferentnitrogensourcecompositions.Theresultsshowedthatinthesinglefactorexperiment,thenitrogensourceswith highermycelialbiomasswerebranandsoybeanpowder.Thevarietywiththehighestmycelialbiomassinthetriangularflaskwasthe DS-2breedofStrophariarugosoannulata,ndthemediumwiththehighestaverage mycelialbiomasswasPDB(potatoglucose)medium.Byculturing theoptimal DS-2bredinatriangularflask,theoptimalrangeforaddingsoybeanpowder was determined tobe 5- (204号 15g⋅L-1 ,and the amount of bran added was determined to be 0-10g⋅L-1 respectively. On this basis,potato and glucose were used as carbonsources,ndafourfactorthree-levelorthogonal experimentaldesignwasconductedtoobtaintheoptimalformula,whichwas tested and proved to be 5g of bran, 15g of soybean flour,10 g of glucose,200 g of potato,1 g of potassium dihydrogen phosphate, 0.5 gof magnesium sulfate,and 1000mL of ultrapure water.The maximum dry mass of mycelium reached 1.32g⋅100mL-1 ,and the averagediameterwaslessthan2mm.Theexplorationof extracelularenzymeactivityshowedthat theoptimal innineformulahad the lowest laccase activity,which was 0.5360U⋅mL-1 ; On the contrary,the formula with the lowest dry mass of mycelial had the highest laccase activity,peaking at 7.195U⋅mL-1 ; There was a significant negative correlation between cellulase activity and biomass, R=- 0.692.Insummary,theoptimal formula for liquid spawnof Strophariarugosoannulatais acompositionof5gof bran,15 g soy powder, 10g glucose and 200g potato slices, KH2PO4 for1 g and MgSO4 O.5 g,in 1 liter ultrapure water. This formula has high dry biomass, lowcost,andsimpleconfiguration.Sincethefomulahasthebovecharacteristics,itcanbeappliedtoliquidspawnseedtankfor further exploration and research.
