沙门氏菌噬菌体多克隆抗体制备及其检测方法

Abstract:Phagesare mainly composed of proteinsand nucleic acids.Whenabacteriophage enters ananimal’s body, it mayberecognizedasanantigen,therebytriggering theproductionofantibodies.Inordertofurtherstudytheproductionof antibodies intheanimalbodyafteroraladministrationof phages,purifiedbroad-spectrum lyticSalmonellphage SCBS was usedas theantigen tochallenge theNew Zealand Great WhiteRabit.The polyclonalantibodiesagainstthe phage SCBS were prepared,andthe indirectELISA method was established.The phageantibodies were detected asfolows:phages werecoated at a concentration of 1×109PFU/mL ,blocked with 2% bovine serum albumin(BSA)for 3 h,the hyperimmune serum ( 1:500 )was incubated for 6O min,then enzyme-labeled secondary antibody( 1:12000 )was incubated for 45min,the absorbance ( OD450 )was measured after the substrate solution containing TMB reacted for 15 min.The established ELISA methoddemonstratedgoodspecificity.TepositiveandnegativethresholdvalueswereO.372andO.310.Theintra-assaycoefficientofvariationranged from 2.30% to 7.11% ,andtheinter-assaycoefficient ofvariation ranged from 1.37% to 9.25%
