虾源肌球蛋白抗原表位的计算机辅助计算

摘要:利用在线软件和线下软件对来源于刀额新对虾(Metapenaeusensis)的过敏原Met el的B细胞抗原表位和T细胞抗原表位进行预测、筛选以及辅助计算,首先,通过uniprot蛋白数据库获取Met el蛋白质氨基酸序列;其次,用ExPASY ProtParam,SignalP-5.0 Server和TMHMM Server v.2.0在线工具对Met el的理化性质、信号肽与跨膜区域进行分析,用PSIPRED在线工具、SOPMA在线工具和DNAstar软件联合预测计算Met el二级结构,用Swiss model在线软件预测计算Met e1三级结构;再次,用DNAStar线下软件和IEDB在线软件综合预测计算线性B细胞表位,用IEDB在线软件预测计算CD4+T细胞表位和CD8+T细胞表位;最后将所得结果进行B细胞表位和T细胞表的筛选,结果表明:Metel蛋白的B细胞表位为15~28,45~49,92~95,125~130,150~153,255~258位氨基酸;T细胞表位为78~84,223~232位氨基酸.

Computer-Aided Calculation of Antigen Epitopeof Tropomyosin in Shrimp

Abstract:Online and offline software were used to predict,screen and assist in the calculation of the B-cell and T-cell epitopes of the allergen Met el from Metapenaeusensis.Firstly,the amino acid sequence of Met el protein was retrieved from the uniprot protein database.Secondly,the physicochemical properties,signalpeptides,andtransmem brane regions of Met el were analyzed by using online tools ExPASY ProtParam,SignalP-5.0 Server,and TMHMM Server v.2.0.The secondary structure of Met el was jointly predicted and calculated by using the PSIPRED online tool,SOPMA online tool,andDNAstarsoftware,while the tertiary structure of Met el was predicted and calculated by using Swiss model online software.Thirdly,the linear B-cell epitopes were comprehensively predicted and calculated by using DNAStar offline software and IEDB onlinesoftware,CD4+T and CD8+T-cell epitopes were predicted and calculated by using the IEDB online software.Finally,the obtained results were screened for B-cell and T-cell epitopes.The results show that the B-cell epitopes of Met el protein are located at amino acids 15-28,45-49,92-95,125-130,150-153 and 255-258,and T-cell epitopes are located at amino acids 78-84 and 223-232.

虾类过敏原是导致食物过敏的一个重要类别,摄入虾可能导致严重的超敏反应.原肌球蛋白(Tropomyosin)是主要的热稳定虾过敏原.Daul等1研究表明,各品种虾的主要过敏原都是一种原肌球蛋白.虾原肌球蛋白是一种肌纤维蛋白,天然含有2个亚基(相对分子质量分别为38500,36600),其比例为1:1,可通过三磷酸腺苷(ATP)诱导的高离子强度组装成纳米纤维[2.虾原肌球蛋白作为主要的食物过敏原已被证明具有极高的热稳定性,并能承受食品加工活动,可见虾原肌球蛋白的致敏性较强.在棕色虾(Penaeus aztecus)中鉴定出13种不同的过敏原,主要有Met el,Penal,Pan s1和Hom al等种类[3.鉴定虾源肌球蛋白抗原中的表位,对去除虾类食品中的过敏原,开发虾源肌球蛋白疫苗,进行免疫诊断和临床治疗等方面具有重要意义,目前有许多方法可用于研究和确认抗原表位,如抗原-单克隆抗体复合物、蛋白水解或化学切割片段方法等.但这些方法大多较复杂,难以执行或效率低下,需对大批潜在的表位候选物进行实验筛选。(剩余7478字)
