Effect of MsTIFYlOa Gene on Cold Resistance of Transgenic Alfalfa Plants

Abstract:Cold stress is one of the important limiting factors affecting the yield and qualityof alfalfaand its popularization in cold areas.Jasmonate(JA)playsan important role in plant growth and development and cold stress response.The objective ofthis study was to investigate the function of MsTIFY1Oa in cold resistance,a key regulatory gene of jasmonic acid pathway in alfalfa.In this study,MsTIFY1Oa transgenic and wild-type Medicago truncatula was used to determine the phenotype,physiological and biochemical, photosynthetic capacity and expression levels of related genes under cold stress.The results showed that the contents of soluble sugar and soluble protein,activities of SOD,POD and CAT,chlorophyll content and photosynthetic index of transgenic MsTIFY1Oa transgenic plants were higher than those of wild-type under cold stress,and the expressions of cold resistance genes,including MtCML10,MtNAC and MtAOC2 were significantly up-regu lated.Cold tolerance was increased in MsTIFY1Oa transgenic plants by increasing the content of osmoregulatory substances,activity of antioxidant enzymes and photosynthetic capacity,and up-regulating the expression of cold resistant genes.
