Abstract :To investigate the mechanismofteciliaassembly inchoanoflagellates,thisstudyfocusedonacolony-forming choanoflagellte,Salpingoecarosetta.Forthefirsttime,theciiaofSalpingoecarosettaweredisassembledviamechanical shearing,anda LiCl(lithiumchloride)-inducedciliaelongationexperimentwasconductedtoinvestigatemechanisms underlyingthedisassembly,regeneration,andassemblyoftheciliaofSalpingoecarosett.IntheLiCl-induced cilielongationexperiment,afinalmolarityof250mmol/Lwasused.Intheciliadisassemblyandregenerationexperiments, the mecanical shearingconditions were determined.Experimentalresults showthattheciliaof Salpingoecarosetta were restored to their original lengths after a mechanical force was applied on them for 120min ;moreover,LiCl-induceda ciliaelongation by more than 60% after 180min of LiCl treatment.Compared with the cold-stress-induced disassembly of the cilia of Salpingoeca roseta,the mechanical shearing methodwasfaster,moreeficient,andsimplerwith higher repeatability.Thisstudyprovidesafundamentalmethodforinvestigatingthemolecular mechanismof theciliaassemblyin Salpingoecarosetta.
