Abstract:Inorder to studytheroleand mechanism of solubledietary fiber inaleviating constipation,this studyutilized 16SrRNA sequencing,gas chromatography-massspectrometry,quantitativepolymerase chainreaction,and immunobloting methodsinamouse model of constipation.Fromvariousaspectsof intestinalmicroecology,suchas the distributionofthegutmicrobiota,short-chainfattacidmetabolites,andtheintestinalepithelialbarier,thisstudyexplored theroleand molecular mechanismof soluble dietaryfiber in aleviating constipation in mice.The aleviating efectof soluble

dietaryfiberon constipation was furtherconfirmedin humantrials.This studyfoundthat soluble dietaryfiber significantly aleviatedthe symptoms of constipation inmice and humans.The mechanisms of action areas folows:increasing the numberof probioticssuchasParabacteroidesand Ligilactobacilus intheintestinal tract of mice;promoting theproduction of short-chain fattacidssuchasaceticacid;and significantly improving theexpresionof genes encoding the production of intestinaltight junctionproteins,alongwithproteins themselves,whichareinvolved inmaintainingtheintegrityof the intestinalbarrier.Theresultsindicate thatsolubledietaryfiber has goodaleviating effctsonconstipationinmiceand humans,whichisachievedthroughthe production of short-chain fatyacids,theimprovementof the gut microbiota,and theenhancementof the intestinal barier.This studyfurther confirms the valueof soluble dietaryfiberforapplicationin constipated populations,providing a scientific basis for intestinal research and product development.
