AbstractObetie]TestablishaneicentadstableISSR-PCRreactionstemaproprateforSraitgosenori.MethdOrnal test design L16(45) was used. The template DNA,primer, Mg2+ ,dNTPs and Taq enzyme that affect the ISSR-PCR reaction system were optimized,andtheptialISSR-PCRsystemforraitiagosenori wasdetermined.ResultTheexperimentalresultssoedthatina 20μL reaction volume,the optimal ISSR-PCR reaction system conditions for Siraitia grosvenorii were 10× Buffer 2.0μL,Mg2+ 1.5 mmol/L (1.2μL ), dNTPs 0.3 mmol/L ( 2.4μL ),Taq enzyme 1.0U ,primer 0.4μLmol/L ( 0.8μL ,and template DNA 75 ng( 1.5μL ).On the premise of optimalreactiooios,salprirsreeexperetsdprirsitaadgularndseleted Finaly,teosteficitaealingtmperatureofpresassletedCoclusion]hissudyproviddaeferecfortesudyofte close relationship between varieties,genetic resources and biological diversity of Siraitia grosvenori.
