Isolation and identification of lipase producing bacteria and optimization of enzyme-producing conditions

Abstract:【Objective】The purposeof this experiment is to screen lipase producing bacteriaand studytheir enzyme producing ability.【Method】Thesoil samples which had been contaminated byoil pollution foralong time,wereenriched and cultured,and the strains were isolatedandscreened by Rhodamine Bplate method.The shaking tableculture with peanutoilas theonlycarbonsourcewas used to explore the enzyme production parametersof theselected strains.The lipase production conditions of each strain weresystematicallyoptimized.【Result】Three strains with high enzyme production ability were obtainedusing Rhodamine B plate methodand namedasLN2,Bk3andBd4.Throughbymorphologicaland16S rRNAmolecular identification,LN2wasPseudomonasaeruginosa,Bk3was Burkholderiavietnamiensisand Bd4 was Acinetobacter soil.After 48 hours of cultivation,the optimal temperature for the three strains is 30~40% ,and the optimal culture pH is 7.O .Through comprehensive screening,the enzyme activity of LN2 was found to be 1.25U/mL at 30% and pH 7.0; the enzyme activity of Bk3 was O.85 U/mL at 40% and pH 7.O; and the enzyme activity of Bd4 was 3.13 U/mL at 30% and pH7.0.【Conclusion】The three lipaseproducing bacteria showed excellent potential in lipaseproduction,especially Bd4, which provided candidate strain materials for the industrial application of lipase.
