Determination of pollen viability and optimization of pollen germination medium in Artocarpus heterophyllusLam.

Abstract:【Objective】This studyaimed to screentheoptimalmedium formulationfor in vitro germinationofArtrocarpus heterophylusLam.pollenandtoevaluate theapplicabilityofrapid staining methodfordetectingpollenviability.【Method] Pollen of‘Siji’A.heterophylus was used as experimental material to measure pollen germinationrateandoptimize polln culture medium composition by single factor test and orthogonal test.【Result】The effects of sucrose, H3BO3 , CaCl2 and MgS04 on pollen germination rate were significant, with CaCl2 ranking first,followed by sucrose, H3BO3 ,and MgSO4 . The optimummedium formulawas 5% sucros Ω3+150mg/LH3BO3+100mg/LCaCl2+150mg/LMgS04 ,and the germination rate was72.39%,which was significantly higher than other treatments.The optimum germination temperature was 27% : Both theMTTstaining method(10 min staining,62.99%viability)andthe TTC staining method(45 min staining, 78.58% (20 viability)couldbeusedforrapid detectionof pollen viability.The MTTstaining method was more eficientand had better contrast ratio while the magenta acetate and I2 -KI methods were not applicable.【Conclusion】The results provide a theoreticalbasisandpracticalsupportforpollenviabilitydeterminationandartificialpolination technology inA. heterophyllus.crossbreeding.
