Abstract:Elevated levelsof hypochlorousacid(HOCl)may beoneof the hallmarks ofchronic inflammatory joint disease.Therefore,thedevelopmentofeffective methods forrapidandaccurate detectionormonitoringofHOClisimportant forfurtherunderstanding theroleofHOClinrheumatoidarthritis(RA).Herein,anear-infrared(NIR)fluorescentprobe (HN)forthe detectionand monitoring of HOCl was reported.In the presenceof HOCl,the phenolic hydroxylgroupof HN is oxidized to produce quinone compounds,resulting inquenching of its NIR fluorescence.Probe HN has several advantages such as long emission wavelength ( λem =660 nm),large Stokes shift(2OO nm),high selectivity and sensitivity (LODis 54.8 nmol/L),and widepHapplicabilityrange(4.5-10.5).The“naked-eye”detectionof HOClinreal water samples has beenrealizedusing the probeHN-basedtest strip.Inaddition,the probe has beensuccessfullyused forfluorescence imaging of exogenous and endogenous HOClin living zebrafish and mice.Most importantly,the probe HN has shown remarkable efectiveness in monitoring HOCl-mediated responses toRA therapy.Thus,HN provides anovelapproach to further understand the role of HOClinRAand evaluate the effcacy of drug therapy for RA.
