褪黑素抗慢性束缚应激致小鼠脑损伤研究

Abstract:Pscholoicalstresisoneofthemaorhealththreatsfacedb com anionanimalssuchasdos andcatsin moderndomesticenvironments.Lon-termexosuretohih-stres conditionscanleadto behavioralabnormalities,supresionofimmunefunction,andneurodegenerativediseases,seriously afectinganimalwelfareandqualityoflife.ThisstudyestablishedapsychologicalmodelinKunmingmice through28daysofchronicrestraintstres andthenintervenedwithexogenousmelatonin.Behavioral, ELISA, biochemical, histological and immunohistochemical techniques were used to evaluate the potential protectiveefectofmelatonin(MT)onbraininjurycausedbychronicrestraintstres (CRS)inmice.The results showed that CRS led to anxiety and depresion-like behaviors in mice, significantly increased the levels of CORT, NE, IL- ⋅1β , IL-6 and TNF- ∇⋅α in the plasma and hipocampus of mice ϕ<0.05) ), significantl decreasedthelevelsof5-HT,MTandDA ⋅p<0. 05⟩ ),andsinificantlincreasedthelevelofMDA inthe lasmaofmiceanddecreasedthelevelsofCAT,SOD,GSH- ⋅Px andT-AOC ).Further studies found that CRS significantly reduced the proportion of positive expresion areas of prefrontal cortex andhipocampusneuronsandNeuNproteininmice ).Incontrast,aftertheinterventionwith MT, the stres response and anxiety and depresion-like behaviors caused by CRS in mice were aleviated, thelevelofoxidativestresinthebod wasinhibited,andtheneuronsinthebrainofmicewas rotected, whichwasmanifestedasasinificantl decreaseinthelevelsofCORT,NE,IL- ,IL-6andTNF- ∇⋅α∇ inthe plasmaofmice ⋅p<0.05) ),anincreaseinthelevelsof5-HT,MT,DAandtheantioxidantenzymes including CAT, SOD and GSH- ⋅Px ${ \it p } < 0 . 0 5 \$ ), thereby inhibiting the production of MDA and the reductionofthepositiveexpresionofNeuNprotein,andaleviatingthedegre ofpsychologicalstres of mice.Inconclusion,thisstud showsthatMThasa otentialtheraeuticefectonthebraininur caused byCRSinmice.
