Construction of a Comprehensive Extreme Waterlogging Tolerant Germplasm Evaluation and Identification System for the Whole Growth Stage of Brassica napus L.

Abstract:Rapeseed (Brassica napus L.) is an important oil crop in China. Waterlogging is one of the most critical limiting factors in rapeseed production. Screening and identifying waterlogging tolerant germplasm resource and developing waterlogging tolerant varieties is an effective way to enhance the waterlogging tolerance of rapesed. Evaluation and identification waterlogging tolerance is the prerequisite for screening waterlogging tolerant germplasm resource and breeding new varieties. Currently,waterlogging tolerance identification is mostly focused on aspecific growth stage with diverse methods,lacking aunified standard.To further improve and standardize the evaluation of waterlogging tolerance in the whole growth stage of rapeseed,indicators and time for waterlogging treatment at different developmental stages were optimized. The waterlogging treatment durations were24 hours,21 days,and 21 days at the germination stage in the laboratory,sedling stage in the field(5-6 leafstage)and flowering stage in the field(fullbloomstage),respectively.Eight rapeseedaccessions were comprehensively evaluated for their waterlogging tolerance using the comprehensive evaluation value calculated by the membership function method,across above three development stages,and one rapeseed accession Zhongshuang 9 was identified with extreme waterlogging tolerance in the whole growth stage. In conclusion,a comprehensive evaluation system for extreme waterlogging tolerance in rapeseed acrossthe whole growth stage was established,which willprovide reliable technical support for the screening of waterlogging tolerant rapeseed germplasm resource,waterlogging tolerance breeding and mechanistic research,as well as offer new ideas for the waterlogging tolerance evaluation in other crops.
