【Abstract】ObjectiveToexplorethe molecular mechanisms of Alarin/NOX1pathway intheprocessesofheartfailure with preserved ejectionfraction(HFpEF)MethodsDalsaltsensitiveratswereusedtocreateanimalmodelsofHpEF.Theratswererandomlydivided intoacontrolgroup,ahighsaltdietgroup,ahighsaltdiet+Alaringroup,ahighsaltdet+overexpresiocontrolgroupaghsaltdie + NOX1 overexpression group,and a high salt die + Alarin + NOX1 overexpression group,with 5 rats in each group. Changes in E/e’and left atrial area ofratsweredetectedbychocardiography.Thelevelsofreactiveoxygenspecies(ROS)and8-hydroxy-2'-deoxyguanosine(8-OHdG)in myocardialtisuewasdetectedbyELISAmethod.HEstainingwasusedtodetectmorphologicalchangesinmyocardialtissue. ImmunohistoccaldetetofarinndNX1protexpresioninmyoadaltss.Masosgsusdtetettheposio of collgefibs inyocadialisse.Teexpressoofryaodinereptor2(Ry)andsarcoplasmicetimcalimpup(SEC2) inmyocardialisseasdetecedbWesteblotResultsThlftatrialareaandE/e’indexsignificantlecreadinteighaltdiet Alarin group and the high sat diet+Alarin + NOX1 overexpression group.The high salt diet+Alarin group and the high salt diet+Alarin + NOX1 overexpressongroupsignificantlyreducedthecontentofROSand8-OHdGinthemyocardialtissueofrats.Thehighsaltdiet + NOX1 overexpressiodsdoalllsedolodallldd muscle fibers.Mason staining results showed that collgen fibers were most significantly increased inthe high salt diet + NOX1 overexpression group.Thehighsaltdiet+laringoupsignificantlyducedteexpresionevelofy2proteinintemyocardialtisueofatsilethe expresionlevelofSERCA2proteinsignificantlyincreased.ConclusionNOX1wassignificantlyincreasedinHFpEFats.NOX1combind with Alarin to improve oxidative stress,thus improving the cardiac function of HFpEF. 【Keywords】 Heart failure with preserved ejection fraction;NADPH oxidase1;Alarin; Oxidative stress
