【AbstractObectiveToobsevethepathologcalchangesofmyocardialtisseunderdiferentdegresofcoronaryatheroseoticlesions,tomeasue theexpressionlevelsofproteinsaoiatedwiththcyclicGMP-AMPstase(cGAS)stimulatorofinteferones (STING)signalingpathwayincoronaryarteries,andtoinvestigatetheroleofcGAS-STIGinthedevelopmentand progresionof coronaryheartdisease.Methods:Eligiblecasesofcoronary heartdiseaseandcontrolcases wereselectedanddivided intocontrol group with normal coronary arteries and grade I,I,I,and IV coronary artery stenosis groups.HE staining was used to observe and evaluate tepathologicalconditionsofcoroaryarteries,Westeblting wasusedtomeasure theexpressionlevelsofdownsrampro teinsoftheGASSTINGpathwayincoronarytisue,ndELISAwasusedtomeasue thelevelsofinflammatorfctorsinoroayissue.AcorrelationanalsisaspeformedtoinvestigatethecorelationoftheexpressonlevelsofdownstreamproteinsoftheGASSTING signalingpathwayandrelatedinflammatoryfactors withthedevelopmentandprogresionofcoronaryheartdisease.Results:Microscopicexaminationshowedthatcomparedwiththecontrolgroup,theotherfourgroupshadvaryingdegesofpathologicalchanges suchas vascular wall thickening and luminal stenosis,with a gradual increaseinthedegree of stenosis from grade I to grade IV coronarylesions.Comparedwith thecontrol group,the grade l,ll, (204 III ,and IV coronary artery stenosis groups had significant increases in the expression levels of downstreamproteins of the cGAS-STING signalingpathwayand related inflammatory factorsincoronary tissue,with a trend of increase from grade I to grade IV coronary lesions.TheexpresionlevelsofdownstreamproteinsoftheGASSTIGsignalingpathwayincoronarytissuewerepositivelyceated with the development and progression of coronary heart disease(cGAS: r=0.927 P<0.001 : -Sting: r=0.889 , P<0.001 p -TBK1: r= 0.910,P<0.001 -IRF3: r=0.936 P<0.001 ;IFN-1: r=0.936 P<0.001 ;TNF- α⋅α∝ r=0.945 , P<0.001 ;IL-1β: r =0.962,P<0.001;IL-6:r= 0.933, P<0.001 ).Conclusion:There are significant differences in the expression levels of downstream proteins of the cGAS-STING signalingpathwayandrelatedinflammatoryfactorsndthissignalingpathwayaybeapotentialargetforthreatmentofooary heart disease.
