【Abstract】 Objective To investigate the role ofdualstaining using resorcinol basic fuchsin for elastic fibers andcytokeratin7（CK7）immunohistochemistryinclarifyingthe grading of pleural invasioninlung adenocarcinoma. MethodsThirty lung adenocarcinoma tisue specimens diagnosed bythe Department of Pathologyat Sun Yat-sen Memorial Hospitalfrom January2024 toJune2O24wereselected.Hematoxylin-eosin（HE）staining，elastic fiber staining，and dual staining combining immunohistochemistry （CK7）with elastic fiber staining were performed.The differencesamong these three methods in determining pleural invasion grading were analyzed.Results Conventional HE staining and elastic fiber staining alone definitively diagnosed pleural invasion grading in 3 cases （ 10% ）.Incontrast，the dual staining method （resorcinol basic fuchsin and CK7）stained tumorcells brown-yellow， elastic fibers blue-black，and nuclei red，enabling 100% definitive diagnosis of pleural invasion grading inall 30 cases.Statistical analysis revealed significant differences among the three staining methods（ χ2=49.091 ， P<0.001 ） ConclusionThe dual staining method with resorcinolbasic fuchsin and CK7 visuallyand clearly demonstrates the relationshipbetween tumorcellsand theelasticlayerof the visceral pleura.It effectively identifies tumorcell invasion into thepleura and facilitates theasessment of pleural invasion grading，providing an'important complementary technique for dual staining inlung adenocarcinoma.
