【Abstract】 Objective To share our insights on the immunohistochemical trichrome staining technique utilized inthediagnosisof Epstein-Barr virus-positivefolliculardendriticcell sarcoma(FDCS).MethodsIn this study,twopatientswithEBV-positiveinflammatory foliculardendriticcellsarcoma(IFDCS)involvingtheliverand spleen,aged 24 and 68 years old respectively,and with tumor diameters ranging from 30 to 55mm ,were selected for immunohistochemical staining ofCD21and CD35,in situ hybridization forEBVencoded mRNA (EBER),and triple staining for CD35,CD21,and EBER,and reviewedthe pertinent literature.ResultsUnder the microscopeof pathological section,the fusiformoroval tumorcells were arranged inschiff-likeor fasciculararrangement.The nuclei of these atypical cells showvesicular chromatin andsmall,distinct nucleolus.The resultsof immunohistochemistry and insitu hybridizationshowedthat CD21and CD35 were positive.EBER in situ hybridization also showed positive results in2cases.The tumor cels were spindle-shaped oroval with a zonal or fascicular arrangement.Theseatypicalcells exhibited vesicular chromatinand small,conspicuous nucleoli.Immunohistochemistryand in situ hybridization revealed that the tumorcels were positive for CD21 and CD35, respectively.EBER in situ hybridization was also positive in both cases.ConclusionStaining for CD21,CD35, and EBER onthe same slide allows for the simultaneous visualization of three outcomes at both the gene and protein levels,enabling a more intuitive analysis of IFDCS tumor cells.
