小麦气孔发育相关基因TaFAMA1A的克隆及初步功能分析

【Abstract】In order to studythe biological function of TaFAMAlA in wheat, the paper made comprehensive analyses usinggenecloning,bioinformaticsapproaches,and quantitativereal-time PCR.Theresults showed thatthe TaFAMAIA gene,containing a coding sequence of 1254 bp and capable of encoding 417 amino acid protein, was successfully cloned from the young leaves of wheat.Bioinformatics analysis showed that the TaFAMA1A protein (molecular formula: C1939H3076N574O635S11) has a predicted molecular weight of 44.9kDa and an isoelectric point of 5.13, classifying it asan unstable protein.The phylogenetic analysis revealed that TaFAMA1A was most closely related to rice FAMA homologous protein.Protein structure prediction indicated thatthesecondary structure ofTaFAMA1Aconsists of a -helix, extension chain, β -foldandrandomcoil,and its tertiarystructurecontainsatypicalhelix-ring-helix (HLH)domain.The quantitative real-time PCR expressonanalysis showedno significantchanges in TaFAMA1A expression underdrought stres,suggesting itmaynotbe directly involved in drought responseregulation.These findings provide afoundation for furtheranalysis of TaFAMA1A in stomatal development or other biological functions in wheat.
