微小RNA-302a-3p靶向溶酶体相关膜蛋白5抑制口腔鳞状细胞癌侵袭转移的研究

[Abstract]ObjectiveThis study aimed to explore the expresionof lysosomal-assciated membrane protein 5 (LAMP5)and microRNA (miR)-302a-3p inoral squamous cellcarcinoma(OSCC)and their functional mechanism on the invasionand metastasis ofOSCC.MethodsThe expressionofLAMP5 in OSCCandits sensitivity asa prognostic indicator were analyzedon thebasis ofThe Cancer GenomeAtlas database.Western blot,quantitativereverse transcriptionpolymerasechainreaction,andcellimmunocytochemistry wereused todetect theexpressionofLAMP5 inOSCC tisuesandcell.TheeffctofLAMP5ontheproliferation,migration,andinvasionofOSCCcellswasevaluated throughcellcountingkit-8,immunocytochemistrymigration,andinvasionassays,respectively.ThemiNAtargeting predictionwebsiteswereusedtopredictthe miRthatregulates LAMP5andverifythe targetedregulatoryefectof miR302a-3pon LAMP5.The effectofLAMP5 knockdownonOSCC tumor growth was evaluated inanude mouse tumorigenesis model.ResultsLAMP5was highlyexpressed inOSCC tisuesandcels.It showed high sensitivity intheearly diagnosisofOSCC.LAMP5knockdownsignificantly inhibitedtheproliferation,migration,and invasionofOSCCcells, whereasLAMP5overexpression increased thesecellactivities.TheexpressionofLAMP5wasregulated by miR-302a3p.In vivo,LAMP5knockdownsignificantly inhibited thegrowthofOSCCtumor.ConclusionLAMP5promotes the malignant progressionofOSCCbyenhancing theproliferation,migration,and invasionofOSCCcels.Theexpression ofLAMP5 is negatively regulated by miR-302a-3p.
