Identification of High Tannase-producing Aspergillus niger Strains and Optimization of Solid-state Fermentation Conditions for Enzyme Production

Abstract:This studyisolatesand screensa fungal strainfromtealeavesusingatannase identification medium.The strain is identifiedasAspergillusnigerthroughmorphologicalcharacteristicsandITSsequenceanalysis,anddesignatedasstrain ALF1-1.FermentedwiththesporesuspensionofstrainALF1-1,thetanaseproductionactivityofblack tealeafstalks reaches 10.42U/mL .Subsequently,the optimal fermentation conditions are determined through one-factor and response surface experiments,which are as follows: temperature 31∘C ,incubation time ,inoculum size 4% ,andammonium chloride concentration 7% .Under these conditions,the predicted enzyme activity is 24.9U/mL ,while the measured activity reaches 25.92U/mL .Compared to the pre-optimization enzyme activity (10.4U/mL ),the optimized solid-state fermentation conditions enhance tannase production by strain ALF11-1 approximately 2.49-fold.
