Abstract：The hematopoietic system combines "transport"and "production" functions，governingthe circulatoryflowof oxygenandcarbondioxide，as wellasthetransport ofnutrientsandmetabolic waste.Oncedamaged，itcan induceanemia， granulocytopenia，acute leukemia，or thrombotic diseases.DOCK family proteins preciselyactivateRac/Cdc42 through signaling axes such as Rho GTPase，BCR，CXCR4，and VEGF，regulating hematopoietic cell chemotaxis，adhesion， andimmunesyapseformation，therebymaintaining immunecellproliferation，differentiation，ndorganismalhomeostasis. Functional defects in DOCK family proteins lead to hematological system disease such as combined immunodeficiency， abnormalredbloodcellmorphology，andrecurrent infections.Elucidating the mechanismofactionoftheDOCK familyin bloodcells notonlyprovides anewperspectivefor thetheoryof hematological systemdisease pathogenesis butalsoopens uppotential strategies forclinical prevention and treatment.
