羊栖菜多酚抗氧化及细胞损伤保护作用研究

Abstract:Objective:Exploring theinvitroantioxidantactivityof polyphenols from Sargassum fusiformeand their protective effects against H22 -induced damage in human aortic endothelial cells(HAECs),thereby providinga theoretical basisfortheirapplicationanddevelopmentinfunctional foodandpharmaceutical fields.Methods : Polyphenols from Sargassum fusiforme were extracted from Sargassum fusiforme with 60 % ethanol,followed by defatted with petroleumetherandpurificationviasilicagelcolumnchromatography.Theinvitroantioxidantcapacitywasassessed by measuring scavenging activitiesagainst1,1-diphenyl-2-picrylhydrazyl(DPPH) radicals,superoxide anion( J2- ) radicals,and 2,2′ -azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS+)radicals.An oxidative damage model was established in HAECs induced by H202 toevaluate the effects ofpolyphenols from Sargassum fusiforme oncell viability andanalyze the dose-response relationship.Results:Thepolyphenols from Sargassum fusiforme showed significant scavenging ability to all three free radicals,and its half-maximal inhibitory concentration( IC50 )was:DPPH radical (204号 0.074mg*mL-1 , O2- radical 0.069mg∙mL-1 ,ABTS+radical 0. 089mg⋅mL-1 .Incellular experiments,polyphenols from Sargassum fusiformealleviated H2O2 -induced oxidative damage in HAECsin a concentration-dependent mannerand significantlyenhancedcell viability( P<0.05 ).Conclusion:Thepolyphenols fromSargassum fusiforme exhibit potent in vitroantioxidantactivityandobviouscytoprotection.Theyactasnaturalantioxidants forfunctional foodsorbiomedical applications to prevent oxidative stress-related diseases.
