Preparation of Oxidized Hyaluronic Acid Crosslinked Carbohydrazide-Gelatin/Cysteine-Chitosan Composite Hydrogel With pH-Responsive and Antioxidant Properties

Abstract:To enhance the biological functionality of natural polymer hydrogels,a pH-responsive composite hydrogel was designed and constructed based on oxidized hyaluronic acid(OHA),cysteine-grafted chitosan (CSCys),and carbonyl hydrazide-modified gelatin(Gel-CDH).The hydrogel achieved dynamic covalent crosslinking through Schiff base reactions between aldehyde groups and amino/hydrazide groups to form a stable and tunable thre-dimensional network structure. By adjusting the OHA ratio,three types of hydrogels were prepared,and their swelling behavior in relation to pH values as well as antioxidant activity were systematically evaluated. The results showed that this composite hydrogel not only exhibited excellnt pH responsivenessbut also demonstrated superior free radical scavenging capability due to the synergistic effects offunctional groups such as thiol groups.
