Abstract:【Objective】Internode length isa crucial trait forbamboo industrial utilization.Pseudosasa japonica var. tsutsumianaisastabledwarfmutant of P . japonica,characterizedbyshortened,flask-shaped internodes,makingitan idealmaterialforstudying internode growth inbamboos.PreliminarystudiesidentifiedtheGRAS transcription factor PjGRAS56 as a key candidate gene responsible for the shortened internode formation in P . japonica var. tsutsumianais. This studyaims to elucidate the potential roleofPjGRAS56 inbamboo internodedevelopment.【Method】Weemployed bioinformatics toanalyze the sequencefeatures ofPjGRAS56,confocal laser scanning microscopy toobserve its subcelularlocalzation,ayeast tanscritionsystemtoassess itstransciptionalactivationactivity,transgenictechques in rice and Arabidopsis to analyze phenotypes of PjGRAS56-overexpressing plants,transcriptome sequencing andqRTPCR to examine gene expression changes,andhand-sectioning to measure internode cellength.【Result】PjGRAS56 localized tothe nucleusand cytoplasm,andlacked transcriptionalactivationactivity.It wassignificantlydown-regulated in the division zone of P .japonica var. tsutsumianais internodes.Overexpression of PjGRAS56 in rice resulted in dwarfism,shortened internodes,reducedtillering,shorter paniclesandawns,fewer grains,andreduced leaf angle; overexpressionof itinArabidopsisledtosmallerleavesandshorterflowerstalks.Althoughthecellengthininternodesof

PjGRAS56-overexpresing plants was similar to controls,thecell number wassignificantlyreduced. Transcriptome sequencinganddiferential gene expresionanalysisrevealedsignificant down-regulationofcelldivision-relatedgenes in PjGRAS56-overexpressing plants.【Conclusion】PjGRAS56isaputative negativeregulatorof internode growth inbamboo.
